Anti-obesity effects of glabridin-rich supercritical carbon dioxide extract of licorice in high-fat-fed obese mice.

Licorice (Glycyrrhiza glabra Linne) is a well-known medicinal plant and glabridin is an isoflavan isolated from licorice. In this study, we investigated the anti-obesity effect of glabridin and glabridin-rich supercritical fluid extract of licorice (LSC). Glabridin effectively inhibited adipogenesis in 3T3-L1 cells. Moreover, LSC showed inhibitory effect on adipogenesis in a dose-dependent manner. The inhibitory effect of LSC resulted from inhibiting the induction of the transcriptional factors CCAAT enhancer binding protein alpha and peroxisome proliferator-activated receptor gamma. Then we fed mice with high-fat diet containing none, 0.1% and 0.25% LSC for 8weeks to explore the anti-obesity effect of LSC in vivo. LSC significantly reduced weight gain by high-fat diet in a dose-dependent manner. The reductions of the hypertrophy of white adipose tissue and of fat cell size were also observed. In the liver, LSC supplementation effectively inhibited high-fat diet-induced hepatic steatosis through downregulation of gluconeogenesis related phosphoenolpyruvate carboxykinase and glucose 6-phosphatase and upregulation of the ╬▓-oxidation related carnitine palmitoyltransferase 1. Taken together, our results suggest that glabridin and glabridin-rich licorice extract would be effective anti-obesity agents.

Lemnul dulce (Glycyrrhiza glabra Linne) este o plant─â medicinal─â bine-cunoscut─â, iar glabridina este un izoflavan izolat din lemn dulce. ├Än cadrul acestui studiu, am investigat efectul de combatere a obezit─â┼úii a glabridinei, ┼či a extractului de lemn dulce bogat ├«n glabridin─â, ob┼úinut prin extrac┼úie cu fluid supercritic (LSC). Glabridina a inhibat eficient adipogeneza ├«n celulele 3T3-L1. ├Än plus, LSC a avut un efect inhibitor asupra adipogenezei, ├«ntr-o manier─â dependent─â de doz─â. Efectul inhibitor al LSC a rezultat din inhibarea inducerii factorilor de transcriere CCAAT stimulatori ai proteinei de leg─âtur─â alfa ┼či a receptorului gama activ la proliferarea peroxizomilor. Apoi am hr─ânit ╚Öoarecii urm├ónd un regim cu con╚Ťinut ridicat de gr─âsimi, con╚Ťin├ónd nici un procent, 0,1% ╚Öi 0,25% de LSC timp de opt s─âpt─âm├óni, ├«n scopul de a descoperi efectul de combatere a obezit─â╚Ťii al LSC in vivo. LSC a redus ├«n mod semnificativ cre╚Öterea ├«n greutate ca urmare a regimului alimentar bogat ├«n gr─âsimi, ├«ntr-o manier─â dependent─â de doz─â. A mai fost observat─â ╚Öi reducerea hipertrofiei ╚Ťesutului adipos alb ╚Öi a dimensiunii celulelor. La nivelul ficatului, suplimentarea cu LSC a inhibat ├«n mod eficient steatoza hepatic─â indus─â de regimul alimentar cu con╚Ťinut ridicat de  gr─âsimi, prin moderarea fosfoenolpiruvat-carboxikinazei ╚Öi glucozo-6-fosfatazei  legate de gluconeogenez─â ╚Öi  stimularea carnitin-palmitoil-transferazei 1 legate de ╬▓-oxidare. Rezum├ónd, rezultatele noastre sugereaz─â c─â glabridina ╚Öi extractul de lemn dulce bogat ├«n glabridin─â ar fi agen╚Ťi eficien╚Ťi de combatere a obezit─â╚Ťii.
